The wild-type amino acid sequence is given along the abscissa. The total number of nonsynonymous mutations for that site in the selected population are indicated over the red bars. In this context 'nonrandom' indicates the alternate hypothesis while 'random' indicates expectations under the null. Note that the M1 start-codon is always preserved as an artifact of ligation during cloning. Thus M1 is a positive control for both unselected and selected pools. Some codon sites, such as E83, exhibited evidence of selection in the unselected pool thereby indicating that the evidence of selection may be attributed to putative cloning artifacts [20].
